Eli Lilly's Jaypirca Cuts Disease Progression Risk by 45% in Previously Treated CLL/SLL (Ref)
Eli Lilly announced positive Phase 3 BRUIN CLL-322 results for Jaypirca (pirtobrutinib) combined with venetoclax and rituximab in relapsed/refractory CLL/SLL, meeting its primary endpoint with a statistically significant 45% reduction in disease progression or death compared to venetoclax + rituximab alone (HR=0.55; p=0.0001)
Median progression-free survival (PFS) was not reached in the Jaypirca arm vs 39.7 months in control. Benefits were consistent across subgroups, including high-risk patients (e.g., TP53 mutation, 17p deletion)
This marks the first Phase 3 study to demonstrate superiority over a venetoclax-containing control arm in CLL, with results applicable to the modern treatment landscape where most patients had prior covalent BTK inhibitor exposure
Lilly plans regulatory submissions to expand Jaypirca’s label and continues multiple Phase 3 studies in CLL/SLL
